What is Lucence Cancer Screening?

Lucence cancer screening is a multi-cancer early detection test that screens up to 50 cancers with a single blood draw. Developed by a team of oncologists and researchers in Singapore, Lucence has gained worldwide recognition, partnering with globally acclaimed laboratory testing healthcare organisations, including Mayo Clinic Laboratories and Agilus Diagnostics.

HOW IT WORKS

The Technology Behind Lucence

Our genes carry instructions for cells to function and grow. When a gene mutates, our growth functions become disrupted. Over time, an accumulation of genetic mutations can turn healthy cells into cancerous cells, potentially leading to development of cancer. As cancer develops and tumour cells grow, these malignant tumours shed fragments of DNA into the blood, known as circulating tumour DNA (ctDNA).

ctDNA contains evidence of gene alterations, containing clues that are characteristic of the organ which they originated. Lucence’s incredibly sensitive amplicon-based NGS technology allows for tiny traces of DNA fragments in the blood to be magnified and studied for mutations that might be present. Leveraging on machine learning to match against extensive cancer bases, Lucence testing is able to pinpoint the location of the tumour with low error rates.

How is Lucence Cancer Screening Different from Tumour Markers?

Traditional tumour markers test for certain proteins found in the blood that may be produced by cancer cells. Common proteins tested in tumour markers include PSA (a protein used to screen prostate cancer), CA-125 (ovarian cancer) and CEA (colorectal cancer).

However, these proteins are not exclusively produced only when one has cancer. Sometimes, these protein levels can be temporarily elevated and can be produced by normal cells when there are hormonal changes, infections or inflammation in the body.

Tumour markers often lack specificity (ability to identify true negatives) and sensitivity (ability to identify true positives), and it is not uncommon for tumour marker test to produce false-positive or false negative results. For example, PSA tumour marker that is used in prostate cancer screening has a specificity of approximately 60% and sensitivity of 72%, reflecting limited precision in detecting prostate cancer as well as a high false positive rate.

  • False negative
    A false negative result refers to when an individual has cancer but their tumour marker reads within the normal range. This happens because early-stage cancers or certain tumours sometimes do not secrete high enough protein levels to be picked up in a tumour marker blood test.
  • False positive
    A false positive result refers to when an individual’s tumour marker test shows elevated levels of proteins when the individual does not actually have cancer. This happens because the proteins are not exclusively produced by cancer cells and can be elevated due to various different factors, such as inflammation, hormonal shifts or infections in the body.

Unlike traditional tumour marker tests that measure protein levels, Lucence tests for ctDNA present in the blood, that are shed by malignant tumours. The test offers a high specificity (measure of how well a test can identify true negatives) of 99% and sensitivity (measure of how well a test can identify true positives) of 81%.

The issue is that not all cancers have biomarkers to measure for, for example, in brain cancer or kidney cancer. This means that cancers not cover under traditional cancer screening have higher chances of going undetected until its in its late stage, with late detection affecting the prognosis and 5-year survival rate.
